CC -!- FUNCTION: Transcription factor that plays an essential role in both
CC trophoblast giant cell differentiation and in cardiac morphogenesis
CC (PubMed:9500551, PubMed:16759287). Binds the DNA sequence 5'-NRTCTG-3'
CC (non-canonical E-box) (PubMed:7791788). Acts as a transcriptional
CC repressor of SOX15 (PubMed:16759287). In the adult, could be required
CC for ongoing expression of cardiac-specific genes (By similarity).
CC {ECO:0000250|UniProtKB:O96004, ECO:0000269|PubMed:16759287,
CC ECO:0000269|PubMed:7791788, ECO:0000269|PubMed:9500551}.
CC -!- SUBUNIT: Efficient DNA binding requires dimerization with another bHLH
CC protein. Forms homodimers and heterodimers with TCF3 gene products E12
CC and E47, HAND2 and HEY1, HEY2 and HEYL (hairy-related transcription
CC factors). Interacts with MDFIC. Interacts with SOX15; the interaction
CC enhances HAND1-induced differentiation of trophoblast giant cells
CC (PubMed:16759287). {ECO:0000269|PubMed:10924525,
CC ECO:0000269|PubMed:16759287, ECO:0000269|PubMed:17891141}.
CC -!- INTERACTION:

CC       Q64279; Q61039: Hand2; NbExp=3; IntAct=EBI-81361, EBI-81388;
CC       Q64279; P15806: Tcf3; NbExp=2; IntAct=EBI-81361, EBI-81370;
CC -!- SUBCELLULAR LOCATION: Nucleus, nucleoplasm
CC {ECO:0000269|PubMed:17891141}. Nucleus, nucleolus
CC {ECO:0000269|PubMed:17891141}. Note=Interaction with MDFIC sequesters
CC it into the nucleolus, preventing the transcription factor activity.
CC Phosphorylation by PLK4 disrupts the interaction with MDFIC and
CC releases it from the nucleolus, leading to transcription factor
CC activity.
CC -!- TISSUE SPECIFICITY: Smooth muscle cells of the gut and adrenal tissue.
CC -!- DEVELOPMENTAL STAGE: Present as a maternal transcript in the egg as
CC well as during cleavage development before blastocyst formation. At 7.5
CC dpc, strongly expressed in all trophoblast cells (PubMed:16759287).
CC Expression seen in the ectoplacental cone and extraembryonic mesodermal
CC components of the amnion, allantois and visceral yolk sac. This high
CC extraembryonic expression persists in the embryonic component of the
CC placenta throughout development. In the embryo, expressed at 7.75 dpc
CC in the lateral mesoderm along the entire length of the embryo as well
CC as throughout the precardiogenic mesoderm. At 8.0 dpc, in the
CC developing heart, expression becomes restricted to the rostral and
CC caudal regions of the straight heart tube, which are fated to form the
CC conotruncus and left ventricle, respectively. Symmetric expression is
CC observed along the left-right axis in the caudal heart tube and lateral
CC mesoderm. As cardiac looping occurs, the interrupted anterior-posterior
CC patterning is maintained with expression in the future left, but not
CC right ventricle. Expressed in the myocardium, but not in the
CC endocardium, and specifically on the greater curvature of the looping
CC heart which is opposed to the pericardium. After day 10.5 dpc, the high
CC cardiac expression level declines abruptly. By 13.5 dpc, expression in
CC the heart is restricted to the regions of valve formation. Besides the
CC heart, expression becomes detectable in the gut at 9.0 dpc. At 10.0
CC dpc, expressed also in the lateral mesoderm and in the neural crest-
CC derived branchial arches. At 10.5 dpc prominent expression in the gut,
CC pharyngeal arches and in sympathetic ganglion primordia. At that stage,
CC a low level of transient expression is seen in the distal posterior
CC region of the limb bud. At 12 dpc expressed in the conceptus
CC trophoblast giant cell layer in the placenta (PubMed:16759287). At 13.5
CC dpc expressed in neural crest derivatives, with abundant expression in
CC the autonomic nervous system and adrenal medulla.
CC {ECO:0000269|PubMed:16759287, ECO:0000269|PubMed:7649392,
CC ECO:0000269|PubMed:9500551}.
CC -!- PTM: Phosphorylation by PLK4 disrupts the interaction with MDFIC and
CC leads to translocation into the nucleoplasm, allowing dimerization and
CC transcription factor activity. {ECO:0000269|PubMed:17891141}.
