背景信息：

乳腺癌易感蛋白 BRCA1 和 BRCA2 在遗传性乳腺癌和卵巢癌病人体内经常会发生突变，并且在与 DNA 损伤，修复，细胞周期进程，转录，泛素化和细胞凋亡等相关的多个过程中都能发挥作用 (1-4)。据悉，在 Rad51 定位到双链损伤 (DSB) 区域时 BRCA2 必不可少，并且细胞缺乏 BRCA1 和 BRCA2 就不能通过 Rad51 依赖的同源重组过程 (HR) 修复 DSB (5)。据观察，已经在 BRCA1 蛋白上发现了多个 DNA 损伤诱导的磷酸化位点，其中包括 Ser988、1189、1387、1423、1457、1524和1542，并且依赖于细胞周期的方式激活的激酶，包括 Aurora A 和 CDK2，可分别磷酸化 BRCA1 的 308 和 1497 位丝氨酸 (6-10)。由 CDKs 催化的细胞周期依赖的 BRCA2 3291 位丝氨酸的磷酸化被认为是 BRCA2 阻止 HR 的机制，因为细胞在前进至 S 期时羧基末端的 Rad51 结合区域被屏蔽 (11)。
在非洲爪蟾蜍中，ATR 依赖和 Claspin 介导的 BRCA1 募集导致 Ser1524 (12) 磷酸化，作为对 DNA 损伤作出的应答。
